Tumor metastasis is a complex biological process co-regulated by multiple genes, multiple steps, and multiple pathways, involving a series of pathological changes such as tumor cell invasion, migration, colonization, and immune escape. With the rapid development of molecular biology and genome sequencing technology, a large number of driver genes and suppressor genes related to cancer metastasis have been identified, and their regulatory networks and mechanisms have been gradually elucidated. Breakthroughs in gene therapy technology based on targeted metastatic genes provide a new direction for the precise treatment of advanced metastatic cancer. This article systematically reviews the classification and mechanism of key genes related to cancer metastasis, summarizes the research progress of mainstream gene therapy technologies such as CRISPR gene editing, gene silencing, and targeted gene delivery, analyzes the bottlenecks and limitations of current clinical applications, and looks forward to the future development trend of metastatic cancer gene therapy, aiming to provide theoretical reference for the research of tumor metastasis mechanism and the development of new treatment options.
